Nano particle size influences nano drug delivery systems for targeted drug release

The dosage of pharmaceuticals like cytostatic drugs is relatively low in traditional pharmaceutics because of their strong side effects. Modern therapeutic systems can now apply these drugs more efficiently. Such drug delivery systems are constructed technologies for controlled drug release and delivery at the targeted area of the body. Development of application systems, which offer improved therapeutic effects at a higher therapeutic security while avoiding inconvenient side effects, is the subject of countless research projects.

Particle size distribution of dextran nano particles

Many new drug candidates for cancer therapy, neurodegenerative diseases or infectious diseases face various obstacles on their way of becoming a pharmaceutical, e.g. release, solubility, distribution and transport, residence time, metabolism and toxicity. Advanced dosage forms are the essential components of strategies to overcome these obstacles.

Modern drug release systems are based on liposomes, dextran nanoparticles, microspheres, gels, prodrugs or cyclodextrins. Particle synthesis as well as drug-loading affects the particle size of the drug release system. Drug transport, release and cell absorption are directly related to particle size. To control a successful construction of a carrier system and a successful drug charge, sensitive measuring systems are necessary which allow a reliable particle size analysis within nanometer range.

Based on the principle of dynamic light scattering (DLS), NANOPHOX uses photon cross-correlation spectroscopy (PCCS) for particle size analysis in a wide concentration range. Successful drug charge of dextran nano particles with the ingredient doxorubicin is documented by a mean particle diameter increase of 37 nm to 45 nm. Furthermore, NANOPHOX enables the in-situ-observation of drug release depending on pH value or stability of system over time.
